I just purchased the 1661 (w/ $100.00 airtime) and a new Rogers phone to use the sim card in. Do I need to activate the same card in the 1661 or can I activate it with it in my new phone? Does it even matter what phone the sim card is in when I call in to activate?

No, it doesn't matter what phone the sim card is in. It doesn't even have to be in any phone, since you have to activate it from a separate phone anyway.

Indeed. Speak Out sim cards will worked in locked Rogers phones.
Getting Rogers to unlock a purchased phone is another story in frustration entirely.
